What is the background of the project?
Please, provide the background of the project (Provide information about the project; min. 10 lines, max. 25 lines) Due to increasing antibiotic resistance among pathogenic bacteria and inefficient strategies to combat biofilm infections, rigorous search for new alternative antimicrobial and anti-biofilm agents are subjects of many recent studies. In this regard, in recent years, phage endolysins have been investigated as potential antimicrobial agents that kill the cells by disrupting the bacterial cell wall.The Turkish Society of Microbiology, founded in 1882, organizes seminars and conferences that aim to educate microbiologists on newly emerging bacteria and bacteriophage identification methods.The Turkish Society of Microbiology have many different group ;Forensic Medicine Microbiology Working Group,Anaerop Working Group,Standardization of Antibiotic Susceptibility Tests (ADTS) Working Group,Food and Waterborne Pathogens Working Group .
What is the aim of the project?
This project is aim to develop a new anti-staphylococcal recombinant endolysin isolated from lysogenic bacteriophages, which were obtained from clinical strains of S. Aureus.
What techniques and methods are used?
-Amplification of endolysin gene from phage DNA by PCR (Polymerase chain reaction)-it is a method widely used in molecular biology to make several copies of a specific DNA segment -Southern blot is a method used in molecular biology for detection of a specific DNA sequence in DNA samples. Southern blotting combines transfer of electrophoresis-separated DNA fragments to a filter membrane and subsequent fragment detection by probe hybridization. -Cloning and expression of endolysin genes with nothern blot. The northern blot, or RNA blot is a technique used in molecular biology research to study gene expression by detection of RNA (or isolated mRNA) in a sample. -Statistical Package for the Social Sciences (SPSS) 26, statistical analysis, data management (case selection, file reshaping, creating derived data) and data documentation (a metadata dictionary is stored in the datafile) are features of the base software.

What are the tasks expected to be accomplished by the student?
What are the tasks expected to be accomplished by the student? (Minimum 10 lines, provide detailed explanation) The following tasks will be expected to be accomplished by the student: -Learning microorganisms that cause disease in human and microorganisms’ working mechanisms - Examining the steps of DNA (deoxyribonucleic acid) isolation which will be done by the tutor. - - Usage and applying of the PCR method with the help of the tutor (polymerase chain reaction) which is a method widely used in molecular biology to make several copies of a specific DNA segment amplifications. - Following steps of cloning and expression of endolysin genes recombinant endolysin isolated from lysogenic bacteriophages, -Observing Biological activity test,recognizing bacterial activities such as in vitro conditions.
Will there be any theoretical teaching provided (preliminary readings, lectures, courses, seminars etc)
There are many different subjects open for understanding methods.The subject will be taught once per week by tutor Prof. Dr. Ali Osman KILIC, depending on the availability.. -Bacteria gene transfer mechanism has 3 main step: transformations,conjugation,transduction -Principles of Polymerase Chain Reaction (PCR) -Functional Analysis Methods(Mutation, Immunoblotting with peptides, Plasmid Purification) -Steps of cloning are explained isolated from lysogenic bacteriophages,
