What is the background of the project?
Pancreatic cancer is one of the most aggressive human oncological disease, characterized with early metastasis and very poor survival . Neural (perineural) invasion is considered of one the hallmarks of pancreatic cancer , which is linked to tumor dissemination and local recurrence of surgically resected tumour.. Therefore, neural invasion of tumor cells is considered as specific root for tumour spread. Our research group developed experimental model of in vitro nerve invasion and established nerve invasion pancreatic cancer cell cloned. By analyzing these clones, deregulation of several - potentially nerve invasion specific genes were found ( such as Macf1, PRR13 etc.)
What is the aim of the project?
Aim of the project is to characterize expression and function of identified genes in pancreatic cancer and in particular to perineural invasion of pancreatic cancer.
What techniques and methods are used?
-Expression analysis of tissues (immunohistochemistry) -Expression analysis of cells (immunoflurescence) -Transient silencing of gene expression in cells -Analysis of cells growth/apoptosis Tools used: Cell culture, cell/tissue staining, microscopy, colorimetric experiments, data analysis.
What is the role of the student?
- The student will observe the practical experiments but will be highly involved in the analysis of the results
What are the tasks expected to be accomplished by the student?
Student will be asked to review literature to gain knowledge about the project. Prepare presentation about topics and write small review. In parallel, student will be allowed to attend lab experiments, such as : cell culture and preservation, gene silencing experiments,immunostaining of cells and human pancreatic tissue. Students will work with microscopic imaging of cell/tissues.
Will there be any theoretical teaching provided (preliminary readings, lectures, courses, seminars etc)
Students will go through theoretical teaching under the supervision of tutor. This includes : Preliminary readings and literature review of topics (6 days) Theoretical teaching of research methods in parallel to lab work (6 days) Teaching of figure design and data analysis.(3 days)
