What is the background of the project?
Regenerative medicine has developed and several clinical applications have been already performed. In addition to stem cell biology, tissue engineering is an essential research fields for regenerative therapy and human tissue model fabrication. Recently, meat production by tissue engineering has been also focused in the world. In this situation, we have engaged in cardiac/skeletal muscle tissue engineering. We have already functionally contractile cardiac/muscle tissues, however the tissues are relatively immature. In this project, we would like to challenge more maturated muscle tissues.
What is the aim of the project?
The main aim of the project is to improve maturation of engineered muscle tissues by some mechanical and/or nutritional treatments. It will contribute to in vivo-like muscle tissue fabrication for future regenerative therapy, model fabrication and meat production.
What techniques and methods are used?
We have already established the culture of cardiomyocytes/myoblast. So, in this project, some mechanical stress induced by electrical stimulation and/or stretch device will be applied to these cells. Additionally nutritional control may be performed for muscle maturation. Engineered constructs will be assessed by histology, immunofluorescent staining and force measurement.
What is the role of the student?
- The student will observe the practical experiments but will be highly involved in the analysis of the results
- The tasks of the student will be performed on his/her own
- The tasks will be done under supervision
What are the tasks expected to be accomplished by the student?
First, the task is successful culturing of cardiac/skeletal muscule cells. Then appropriate condition control regarding electrical stimulation, stretching and nutrition will be discussed and performed. Time course studies will be performed. Contraction power will be measured by the pre-existing measurement system. After tissue fixation, histological analyses will be performed. Immunofluorescent staining for muscle specific marker will be also performed. The data will be analyzed and discussed. Confirmation of the maturation of engineered muscle tissues will be expected.
Will there be any theoretical teaching provided (preliminary readings, lectures, courses, seminars etc)
Our faculties will give several lectures about tissue engineering. The student also will attend the weekly seminar and present his/her work in the last week.
