What is the background of the project?
Recurrent respiratory infections are a prominent feature of Cystic Fibrosis. It is important for testing of antimicrobial susceptibility and antimicrobial strategies for Cystic Fibrosis. To date, considerable scientific experience has been gained in the microbiology of cystic fibrosis.Formed the idea of the main clinically significant pathogens, their epidemiology, the role in the development of pulmonary complications, approaches to microbiological diagnosis.If earlier, before the advent of mass spectrometry and 16S RNA sequencing, our understanding of the microbes associated with the inflammatory process in the lung tissue was reduced to a dozen of the main pathogens, today this number is already striving for a hundred.
What is the aim of the project?
Mastering the method of mass spectrometry in laboratory diagnostics of infectious complications of Cystic Fibrosis. The aim of the study is to analyze the homogeneity of the population of strains isolated from patients with cystic fibrosis from different regions of the Russian Federation through their subtyping, using mass spectrometry. The expected result depends on the level of preparedness of the young scientist, both on theoretical knowledge and on the ability to analyze and structure the results obtained, and can be presented as a publication in a journal reviewed by the Higher Attestation Commission and / or international publications, and also as a poster or oral presentation at conferences at various levels.
What techniques and methods are used?
Microbiological method: cultivation of microorganisms on basic and selective media. Identification of microorganisms MALDI TOF mass spectrometry. Analysis of the results using software FlexAnalysis With MALDI TOF we can detect the particular properties of the isolated specific pathogen, associated with Cystic Fibrosis, identification and determination of the their properties, microorganism structure determination, antibiotic resistance determination, definition of phylogenetic relationship of the isolated microorganisms. Mass spectrometry is a method for investigation a substance based on determining the Mass to charge ratio of ions formed during ionization of the sample components of interest. One of the most powerful ways to qualitatively identify substances, which also allows quantitative determination.
What is the role of the student?
- The student will observe the practical experiments but will be highly involved in the analysis of the results
- The tasks will be done under supervision
What are the tasks expected to be accomplished by the student?
Microbiological examination of clinical material from patients with Cystic Fibrosis (swabs from the back of the throat, sputum): primary bacterial cultures, identification and determination of the properties of the isolated microorganisms, microorganism structure determination, antibiotic resistance determination, definition of phylogenetic relationship of the isolated microorganisms. The researcher will independently carry out the isolation of strains on nutrient media, extract and obtain the protein spectra of the microorganism using mass spectrometry, study the characteristics of the protein profile of these strains, carry out their subtyping, evaluate the results, interpret them, format the data obtained during the experiment. articles or other format.

What skills are required of the student? Is there any special knowledge or a certain level of studies needed?
Knowledge of working with a microscope; ability to work with cultures of microorganisms; knowledge of main diagnostic methods. Subjects passed: Microbiology/Bacteriology.
