What is the background of the project?
We previously identified molecules, which are involved in function of neural stem cells during brain development. Results of our recent experiments suggested that the genes we found are involved not only in neural stem cells but also other aspects of brain development and regeneration and aging multiple organs. Human genome analyses by other research groups suggested that the genes are involved in psychiatric diseases and normal brain functions of human, such as intelligence. However, experimental confirmation of these possible molecular functions has not been reported yet.
What is the aim of the project?
Reveal roles of some genes and their protein products of our interest in regulation of brain development and aging.
What techniques and methods are used?
Histology (basic staining methods and immunostaining). Cell culture experiments (overexpression of proteins of interest, and drug induced gene knockout in the culturing cells). Analysis of protein expression by western blot. Immunoprecipitation assay. Analysis of expression of genes by RT-PR and microarray.
What is the role of the student?
- The student will observe the practical experiments but will be highly involved in the analysis of the results
What are the tasks expected to be accomplished by the student?
Performing some experiments and data analysis. Basic technique of our lab is histology, not only basic staining methods, but also immunostaining and in situ hybridization. Utilizing these histological techniques, we study abnormalities in model mutant mice. To know the function of molecules that we study, we also perform biochemical and cell biological experiments. The student is expected to learn and perform multiple experiments to analyze mutant mice and function of the molecules that is mutated in the mice. When the student gets good results of experiments, he or she is expected to collate the data to make the data usable in paper publication.
Will there be any theoretical teaching provided (preliminary readings, lectures, courses, seminars etc)
Yes. We will teach the detail of the project and our aims, and also suggest papers to read for understanding our researches. Our lab staff are always available, when the student need supervising to carry our experiments. We will provide theoretical teaching at that time.
What is expected from the student at the end of the research exchange? What will be the general outcome of the student?
- No specific outcome is expected
What skills are required of the student? Is there any special knowledge or a certain level of studies needed?
Histology and cell biology. It is not needed to say that perfect skills are not expected. It is enough, if the student is interested in our project.
