What is the background of the project?
The knowledge on the molecular profile of brain metastasis (BM) from lung cancer is relatively scarce. This is partly due to difficult accessibility of the valuable diagnostic material. Indeed, BM occur mainly in patients with advanced disease at presentation, who are not candidates for thoracic surgery, therefore most patients with untreated BM is excluded from recent trials investigating new therapies in lung cancer. Poor prognosis and low availability of pair-matched tumor samples from primary tumor and BM limited studies on tumoral-heterogeneity. However, it was confirmed that molecular subclonality between corresponding cancer lesions may cause low efficacy of systemic therapies in lung cancer patients.
What is the aim of the project?
Characterization of the tumoral-heterogeneity between  primary lung cancer and their paired matched BM.
What techniques and methods are used?
First column-based DNA extraction (an extraction method to purify nucleic acids) will be done. Then preparation of library for next generation sequencing, Sequencing for copy number variations (to detect copy number variants) and determination of exomic mutations (using a exonic splicing enhancers program), and finally computationalanalysis of data, to analyse the results.
What is the role of the student?
- The student will observe the practical experiments but will be highly involved in the analysis of the results
- The tasks of the student will be performed on his/her own
- The tasks will be done under supervision
What are the tasks expected to be accomplished by the student?
The student will be conducting the laboratorial experiments (Column based DNA extraction, Sequencing for copy number variations and scaning of slides on the microscope). Supervision (both experimental and theoretical) will be provided on each step of the work and when conducting the labortorial experiments. It will help to develop a hands-on approach and technical expertise of the student during project progression. Scientific documentation (lab journal, notes, and project summary) necessary from the student.

Previous laboratory experience preferred.
