What is the background of the project?
Migraine is a recurrent unilateral pulsatile headache with a significant negative effect on the quality of life. The clinical manifestations of migraine are nausea, vomiting, photophobia, and phonophobia and sometimes, transient neurologic attacks known as “aura”. Based on the presence or absence of aura, migraine is classified into two particular subtypes, such as migraine without aura (MO) and migraine with aura (MA). Several factors including genetic, environment, hormonal factors, and neurotransmitters play roles in the disease etiology. Among the genetic factors, methylenetetrahydrofolate reductase (MTHFR) gene can be studied in more detail due to its effect on the plasma level of homocysteine. Also, in recent years several studies have suggested that the inflammatory process and the components of the immune system may play an important role in the pathogenesis of migraine.
What is the aim of the project?
This study aims to evaluate the association between clinical aspects of migraine and the genetic polymorphism of cytokines in patients attended at the academic outpatient clinic of PUCPR - Campus Londrina / PR.
What techniques and methods are used?
The universe of this study will be constituted by a convenience sample with 100 participants attended at the academic outpatient clinic of PUCPR campus Londrina, PR between 18 and 60 years old, of both genders. The diagnosis of migraine will be made according to the 3rd International Classification of Headache. 5 ml of blood will be collected by venipuncture and the extraction of leukocyte DNA from the peripheral blood will be performed. The polymorphism of the genes: IL-1α-88934; IL-1β + 395435; -174 of the IL-6 gene, IL-10 -62736 and by PCR-RFLP. Fisher's exact test and the chi-square test will be used to establish significant differences between the allele and genotype frequencies of interleukins and TNFα, sociodemographic and behavioral variables, and associated comorbidities between the groups of patients with and without migraine. The odds ratio will be calculated using the Odds Ratio test with a 95% confidence interval (CI). The significance level of 5% will be adopted.

What are the tasks expected to be accomplished by the student?
The student will observe the practical experiments and “lab work” (molecular genetic laboratory) with the active part in the polymerase chain reaction (PCR) and electrophoresis techniques to genotyping and will the student take active part in the clinical examination for diagnosis of migraine in accordance with the International Headache Society (HIS) criteria. The student will make the statistical analyzes of the results obtained.as well as Allele and genotype frequencies and the Hardy–Weinberg equilibrium that will be calculated by chi-squared test using the SNPStats online software.
Will there be any theoretical teaching provided (preliminary readings, lectures, courses, seminars etc)
The student will be able to attend the classes in the graduation of the medical course and also will be carried out directed studies, seminars and discussion in groups in the laboratory of molecular genetics of the institution.
