What is the background of the project?
Type 2 diabetes is one of the main chronic non-communicable diseases worldwide. Currently, most evidence suggested that vitamin D was involved in type 2 diabetes. Since the activation of inflammatory pathways disrupts proper insulin signaling, it is supposed that vitamin D could influence glucose homeostasis by modulating inflammatory response. However, human studies investigating the impact of vitamin D supplementation on inflammatory biomarkers of type 2 diabetes have generated conflicting results. We hypothesized the conflicting results come from inconsistent vitamin D baseline level in these subjects. So, in this study, all vitamin D deficient elderly type 2 diabetes are selected to study the effects of vitamin D supplementation on inflammatory biomarkers of vitamin D deficient elderly type 2 diabetes.
What is the aim of the project?
To identify the health effects of oral vitamin D in elderly patients with type 2 diabetes mellitus (T2DM). Further research the mechanism of vitamin D intervention on the occurrence and development of diabetes.
What techniques and methods are used?
Community intervention trial, animal experiment and Cell experiment. In this project, there are mainly three stages. First, a community intervention experiment has being performed to identify the health effects of oral vitamin D in elderly patients with type 2 diabetes mellitus (T2DM). And methods of community intervention trial would be fully employed here; Second, the specific cell experiments includes the growth of HUVECs(Human Umbilical Vein Endothelial Cells,HUVEC), migration of HUVECs and cytoskeleton change will been done to clarify the mechanism of vitamin D intervention on the occurrence and development of diabetes. And methods of cell experiments (such as cell culture, cell treatment, transfection, gene knockdown) will been used here; Third, an animal study will been conducted to confirm the intervention on the occurrence and development of diabetes. And the methods of animal experiments (e.g. handle the mice, how to build the model of diabetes, collect the tissues) will be used here.
What is the role of the student?
- The student will observe the practical experiments but will be highly involved in the analysis of the results
- The tasks will be done under supervision
What are the tasks expected to be accomplished by the student?
The student need to complete part of the cell experiments under the supervision of the lab manager independently after he or she is familiar with the environment. There are three steps during 4 weeks. In the 1st week, the student need to learn the methods of cell culture to avoid contamination of our cell culture room and the protocols of cell biology. And in the following 3 weeks, the student will do the specific cell experiment includes the growth of HUVECs, migration of HUVECs, MLC protein expression and cytoskeleton change. At the same time, he needs to help us with data entry and analysis of community intervention experiment with SPSS software.
Will there be any theoretical teaching provided (preliminary readings, lectures, courses, seminars etc)
The Cell culture training and Seminar will be held regularly.
What is expected from the student at the end of the research exchange? What will be the general outcome of the student?
- The student will prepare a presentation

- The student will have the opportunity to present the results together with the supervisor at a conference
