What is the background of the project?
Impaired wound healing remains a common, unmet clinical need. This is of particular relevance to diabetic and oncologic patients, where a poorly healing wound often times leads to complications and may even result in amputation of the affected limb. Successful wound healing requires a burst of inflammation at the injury site. At the same time, unchecked inflammation is the hallmark of poorly-healing wounds. Human alpha-1 antitrypsin (AAT) is an endogenous, serum glycoprotein with established anti-inflammatory and tissue protective properties. We have recently demonstrated that AAT is able to accelerate and enhance closure of acute wounds in several preclinical models suggesting it holds potential for treating chronic wounds as well.
What is the aim of the project?
Preclinical assessment of a novel wound healing treatment for diabetic patients and chemotherapy-treated patients.
What techniques and methods are used?
In-vitro, cell monolayer repair and migration assays are used to dissect specific cellular responses. Response to injury-related stimuli is assessed using ELISA (Enzyme-Linked Immunosorbent Assay) and similar assays. In-vivo, excisional wound models are used to evaluate and optimize the therapy. In addition, skin revascularization is evaluated using live fluorescent imaging (IVIS- in vivo imaging system).
What is the role of the student?

What are the tasks expected to be accomplished by the student?
The student will assist in designing and conducting experiments. Specific duties include: 1. Monitor wound closure rate 2. Perform fluorescent imaging (IVIS) 3. Perform epithelial monolayer repair assays 4. Preform cell migration assays In addition, the student will be required to organize and analyze the acquired data. Finally, the student will assist in drafting and preparing the data for scientific publication.
Will there be any theoretical teaching provided (preliminary readings, lectures, courses, seminars etc)
Online animal care guideline course. Plus reading material, scientific literature: 3-5 review articles.
What is expected from the student at the end of the research exchange? What will be the general outcome of the student?
- The student will prepare a presentation
- The student’s name will be mentioned in a future publication
- The student will have the opportunity to present the results together with the supervisor at a conference
What skills are required of the student? Is there any special knowledge or a certain level of studies needed?
There will be non-invasive small animal work involved (using an imaging system). This is the reason we request that the student(s) obtain an online course prior to arriving. Other than that, we require active brainstorming in lab meetings. Subjects passed: Immunology
