What is the background of the project?
The main focus of the project is the characterisation of the role of the stem cell-associated transcription co-factor, ZNF521, in the control of the homeostasis of the stem/progenitor cell subset in the haematopoietic, neural and osteo-adipogenic systems. In this context, the biological effects of the up- or down-regulation of the expression of ZNF521 in stem and progenitor cells will be investigated, as well as the molecular and functional interactions of this protein with signal transduction pathways that have been implicated in the control of stem cell self-renewal and/or differentiation.
What is the aim of the project?
The aim is to characterise the mechanism of action and molecular interactions of ZNF521 in the control of haematopoietic, neural and mesenchymal stem cells.
What techniques and methods are used?
Techniques • Tissue cultures; • Expression vectors; • Gene expression profiling of ZNF521 in haematopoitetic, neural and adipogenic cell lines; • PCR (Polymerase Chain Reaction); • Western Blotting; • Cell growth assays; • Single-cell clonogenic assay; • Soft agar colony formation assay; • Statistical analyses.
What is the role of the student?
- The student will observe the practical experiments but will be highly involved in the analysis of the results
