What is the background of the project?
Certain proteins are expressed in different cells in a periodical rhythmical sinusoidal manner. A group of proteins have periodical increases/decreases of expression corresponding to the day-night rhythm. By means of immunohistochemistry we will map changes in expression of circadian rhythm-related proteins in colorectal carcinoma in comparison to normal intestinal mucosa.
What is the aim of the project?
The aim of the project is evaluation of changes in expression of circadian rhythm-related proteins in colorectal carcinoma in comparison to normal intestinal mucosa, assesment of their prognostic and predictive value.
What techniques and methods are used?
Used techniques: Routine histology for basic diagnostics ov the disease; histochemistry with identification of mucins production by the tissue (PAS, Alcian blue); immunohistochemistry with different antibodies against circadian rhythm-related proteins; morphometry using computerized two-dimensional image analysis for evaluation of particular protein expression.
