What is the background of the project?
Cardiac adaptations promoted by chronic pressure overload are well described; however its systemic impact early in the disease course is poorly described. Some studies raise the possibility of the heart as a modulator of other organs and tissues through secreted molecules, the cardiokines. The adipose tissue and skeletal muscle are probably targets of the cardiokines released as a response to chronic pressure overload.
What is the aim of the project?
In this study we investigate the effects of left ventricular chronic pressure overload in the right ventricle, as well as in adipose tissue and skeletal muscle.
What techniques and methods are used?
The student will perform extraction and quantification of RNA by real time PCR and analysis of the function of isolated fibers from skeletal muscle. Samples from rats submitted to aortic constriction and control animal will be used
What is the role of the student?
- The student will observe the practical experiments but will be highly involved in the analysis of the results
What are the tasks expected to be accomplished by the student?
The student is expected to be perform RNA extraction, analysis of its integrity by electrophoresis, quantification by real time PCR and critical analysis of the results. In this task samples from myocardium, adipose tissue and skeletal muscle will be used. The evaluation of muscle function is a more difficult technique; however, the student is expected to help in the experiments and to analyze the results obtained.

The underlying concepts of each technique and tissue will also be provided by preliminary readings and lectures.
