What is the background of the project?
Serine proteases are the largest known family of protein-cleaving enzymes, and modulate blood coagulation, apoptosis and inflammation. Of these, cathepsin G (CatG), proteinase 3 (Pr3), and human neutrophil elastase (HNE) are three major neutrophilic serine proteases (NsPs) expressed in neutrophil azurophil granules. In particular, HNE and Pr3 are two major NsPs which play crucial roles in antimicrobial defense with overlapping and potentially redundant substrate specificity. However, recent reports suggest that excess proteolysis which results from an influx of neutrophils into the airways is pivotal in the progressive destruction of lung parenchyma e.g. acute lung injury (ALI), acute respiratory distress syndrome (ARDS), and chronic obstructive pulmonary disease (COPD). Therefore, serine proteases (CatG, Pr3, and HNE) may prove to be targets for the generation of agents in the treatment of neutrophilic inflammatory disease.
What is the aim of the project?
Developing lead compound is a milestone in drug discovery. In addition, botanical products and TCM are recognized as important sources of lead agents. The aim of this project is to export lead components/or lead herbals (extracts) from traditional Chinese medicines.
What techniques and methods are used?
In an attempt to identify serine proteases inhibitors, the extracts of TCMs were prepared and their inhibitory effects on serine proteases activities were also evaluated. Using the bioassay-directed fractionation to prepare or isolate the bioactive fractions or bioactive components. CMC data are essential to maintain the quality of botanical products in manufacturing. In addtion, SOPs (standard operating procedures) and quality control were performed for the extracts preparation by HPLC analysis, and bioactive evaluations.
What is the role of the student?
- The tasks will be done under supervision

- The student will observe the practical experiments but will be highly involved in the analysis of the results

- the student will take active part in the practical aspect of the project
What are the tasks expected to be accomplished by the student?
1. To prepare the extracts and fractions of TCMs (maximum 5 extracts). 2. To evaluate inhibitory effects of extracts and fractions on serine proteases activities. 3. To establish the HPLC analysis method and fingerprints of extracts and fractions of TCMs (minimum one extract or fraction). 4. To establish the SOP for preparation of extracts and fractions.
Will there be any theoretical teaching provided (preliminary readings, lectures, courses, seminars etc)
During the period of student in Taiwan, they are able to join any lectures or courses.
What is expected from the student at the end of the research exchange? What will be the general outcome of the student?
- The student will prepare a scientific report
What skills are required of the student? Is there any special knowledge or a certain level of studies needed?
A scientific report is demanded after the exchange
