What is the background of the project?
Nitric oxide is the main neurotransmitter involved in penile erection. Because serotonin is able to decrease blood pressure in rats due to a mechanism involving nitric oxide, we asked whether serotonin could contribute to erection. We observed that serotonin and the antidepressant Fluoxetine (member of SSRI ? selective serotonin reuptake inhibitors) both induce penile erection in rats. However, no erection could be observed in the second consecutive day of serotonin administration; and signs of serotonin intoxication were found after the third or fourth day of serotonin administration. Our hypothesis is that serotonin looses the ability to induce erection in rats due to a mechanism involving receptor internalization. To further investigate the mechanisms involved in serotonin-induced erection in rats such as whether the inhibition of nitric oxide synthase inhibits serotonin responses; to evaluate the mechanisms by which the erections induced by serotonin are lost in the second consecutive administration of serotonin and how to avoid serotonin intoxication. Quantitative measurements of erections induced by serotonin:in vivo using camera recordings and in vitro by isometric tension recordings of cavernous smooth muscle in isolated organ chambers. The experiments will be performed in the absence and in the presence of drugs that may alter serotonin responses (reuptake inhibitors, metabolism inhibitors, receptor antagonists, nitric oxide synthase inhibitors, etc).
