What is the background of the project?
To study of the role of human papillomavirus (HPV) infection in the development of proliferative lesions, we have developed experimental models consisting of primary human keratinocytes expressing E6 and/or E7 oncoproteins from different HPV genotypes either mucosal or cutaneous genotypes. This system will be exploited to assess the production of proinflammatory molecules such as IL-8, MCP1, COX2, ICAM1 ecc. In collaboration with the Dept. of Dermatology, we are studying a patient suffering from Epidermodysplasia Verruciformis (EV); genetic, immunological and virological analysis has been performed. Primary keratinocytes from the EV patient have been recently obtained and will be used to provide evidence for an aberrant response, e.g. cell proliferation or cytokines production, to UV exposure. The results obtained will clarify the role of UVR in skin carcinogenesis and HPV replication.
