The role of leukocyte derived microparticles in airway inflammation
Universita degli Studi di Pisa
Dipartimento Cardiotoracico, presso Ospedale di Cisanello, Via Paradisa 2, 56126 Pisa (PI)
Prof. Alessandro Celi
Dott. A. Celi, Dr. Tommaso Neri
Type of Research Project
- Basic science
What is the background of the project?
IT HAS BEEN KNOWN FOR SEVERAL YEARS THAT EUKARIOTIC CELLS ARE CAPABLE OF SHEDDING MEMBRANE FRAGMENTS, COMMONLY REFERRED TO AS ECTOSOMES OR MICROPARTICLES (MP). ORIGINALLY CONSIDERED IN VITRO ARTIFACT DEVOID OF PHYSIOLOGIC SIGNIFICANCE, MP ARE RAPIDLY GAINING ATTENTION AS MODULATORS IN NUMEROUS PROCESSES, INCLUDING, AMONG OTHERS, INFLAMMATION AND BLOOD COAGULATION. IT HAS BEEN DEMONSTRATED, FOR EXAMPLE, THAT MP CARRY INTEGRAL MEMBRANE PROTEINS LIKE THE INITIATOR OF BLOOD COAGULATION, TISSUE FACTOR, AND THE CELL-CELL ADHESION MOLECULE ICAM-1.
LEUKOCYTE DERIVED MP ARE CAPABLE OF PROMOTING INFLAMMATION BY ACTIVATING LUNG EPITHELIAL CELLS, BUT THE MECHANISMS UNDERLYING THIS PHENOMENON HAVE NOT BEEN COMPLETELY ELUCIDATED. THE PROJECT INVOLVES THE STUDY OF INTRACELLULAR MECHANISMS OF MP SHEDDING.
What is the aim of the project?
What techniques and methods are used?
The project involves the culture of epithelial cells, the isolation of blood monocytes and their culture, the generation of MP from monocytes, the stimulation of epithelial cells with monocyte-derived MP and the evaluation of the epithelial cell response with immuno-enzymatic assays and molecular biology techniques.
What is the role of the student?
- The student will observe the practical experiments but will be highly involved in the analysis of the results
- The tasks of the student will be performed on his/her own
- The tasks will be done under supervision
What are the tasks expected to be accomplished by the student?
The student will be trained to perform all these steps first hand, and will be able to analyze and discuss the data with the tutor.
Will there be any theoretical teaching provided (preliminary readings, lectures, courses, seminars etc)
What is expected from the student at the end of the research exchange? What will be the general outcome of the student?
- No specific outcome is expected
What skills are required of the student? Is there any special knowledge or a certain level of studies needed?
No special requirements.
Are there any legal limitations in the student’s involvement
Type of students accepted
