What is the background of the project?
I. The aim of this project is to find out why ornithine decarboxylase(ODC) results in immature activity and deprived differentiation of leukemia, which results in abnormal proliferation and differentiation of myeloid progenitor cells. Our hypothesis is that ODC interferes with macrophage-like differentiation and matrix metalloproteinase-9(MMP-9) activation signaling by tumor necrosis factor alpha(TNF-alpha) via NF-alpha. Therefore, we aim to accomplish the following studies to elucidate the molecular mechanisms about how ODC interferes with the MMP-9 expression on macrophage-like differentiation. 1.The roles of polyamine biosynthesis pathway in TPA-induced macrophage-like differentiation in HL-60 and U-937 cells 2.The roles of ODC in MMP-9 expression and activity during macrophage-like differentiation
