What is the background of the project?
Use of ectoenztmes (CD38 and CD157) for prognostic applications and therapeutic targeting in human leukemias and solid tumors.
What is the aim of the project?
Studing: -the role of CD157 as receptor that induces reorganization of the cytoskeleton and significant changes in cell shape.It will also study how that signal mediated by CD157 act through modulation of cytosolic Ca(2+) concentration and how CD157 is involved in adhesion to extracellular proteins and in chemotaxis induced in vitro by Fmlp (formyl-methionyl-leucyl-phenylalanine) -the role of CD38,a negative prognostic marker, in B-cell chronic lynphocytic leukemia(B-CLL) in neoplastic B-cell growth and survival and the role of the up-modulation of C D100,restricted to proliferating cells.Finaly,it will study the role of a set of normal receptors and ligands ruling physiologic pathways in B lymphocytes that becomes detrimental in the context of B-CLL cells -the role of CD38 transmembrane glycoprotein as a receptor in cells of the immune system.
What techniques and methods are used?
In viro adhesion assays under static and under flow conditions. Biochemical analysis on signalling mechanisms. Trojan peptide technology. FPLC protein purification. Cell isolation.
What is the role of the student?
- The student will mainly observe
- The student will observe the practical experiments but will be highly involved in the analysis of the results
- The tasks will be done under supervision
What are the tasks expected to be accomplished by the student?
Ideally, the student should have experience in basic science, especially from the view point of complex experiments. We may provide literature useful to understand the project and the general context.
