What is the background of the project?
The process of repolarization of heart muscle cell membrane potential is related to their special potassium ion channels. Failure of some type of the ion channels frequently leads to unexpected heart failure and death. We hope to analyze distribution of the different types of channels (using immocytological techniques and Wetern blot analysis) and understand the function of the different types of ion channels in electric function of the heart muscle cells (by making use of the RNAi technique). We plan to find correlation - with the hope of future genetic counseling - between ion channel coding genes, their mutations and unexpected heart failure. We are in the process of establishing cultures from normal human heart muscle cells as well as from those taken from people deceased due to unexpected heart failure and analyze ion channel functions in the cultured cells.
